Slay the Spire 2 characters
Slay the Spire 2 has five characters: Ironclad, Silent, Regent, Necrobinder and Defect. Only the Ironclad is open at first; each of the others unlocks after you start one run with the character before it. Silent has the highest win rate right now (32.2%).
Ironclad A
The last soldier of the Ironclads. Crushes foes with sword and flame against his will.

A huntress from outside the Spire. Ready to stab and poison anything in her way.

Regent A
Heir to the Throne of Stars. Wields cosmic powers but his minions do the work.

Necrobinder B
A Spireborn lich who seeks revenge. Calls upon her trusty left hand, [gold]Osty[/gold], in combat.

Defect D
An automaton eternally modifying itself to survive. Deploys Orb technology when it must fight.

How to unlock characters in Slay the Spire 2
The order is fixed: Ironclad → Silent → Regent → Necrobinder → Defect. Starting a run is enough. All sts2 characters share the colorless card pool and the shared relics; each also has its own cards and a few relics only it can find. Start a run with the previous character: Ironclad unlocks Silent, Silent unlocks Regent, Regent unlocks Necrobinder, Necrobinder unlocks Defect. You do not have to win, and abandoning the run right away also counts.
